Dual regulation of the glycogen phosphorylase 2 gene Dictyostelium discoideum: the effects of DIF-1, cAMP, NH3 and adenosine.

Cell differentiation in Dictyostelium results in the formation of two cell types, stalk and spore cells. The stalk cells undergo programmed cell death, whereas spore cells retain viability. The current evidence suggests that stalk cell differentiation is induced by Differentiation Inducing Factor (DIF), while spore cell differentiation occurs in response to cAMP. We have discovered the first de...

Possible cooperation of differential adhesion and chemotaxis in mound formation of Dictyostelium.

In the mound stage of Dictyostelium discoideum, pre-stalk cells sort and form a tip at the apex. How this pattern forms is as yet unknown. A cellular level model allows us to simulate both differential cell adhesion and chemotaxis, to show that with differential adhesion only, pre-stalk cells move to the surface of the mound but form no tip. With chemotaxis driven by an outgoing circular wave o...

Dictyostelium Myb transcription factors function at culmination as activators of ancillary stalk differentiation.

ecmB and mrrA are expressed in the cups that cradle Dictyostelium spore heads, and MybE is necessary for their expression in lower but not upper cup cells. A Myb site within the mrrA promoter is necessary for expression in both cups. Thus, multiple Myb proteins are required for ancillary stalk differentiation.

Stage-specific requirement for myosin II during Dictyostelium development.

Dictyostelium cells that lack a functional myosin II heavy chain are motile and are capable of aggregation, but fail to undergo further multicellular development. We have used a Dictyostelium mutant expressing a cold-sensitive myosin heavy chain to examine the requirement for myosin throughout the course of development. The loss of myosin function upon cooling is rapid and reversible. Temperatu...

Glycogen synthase kinase 3 regulates cell fate in dictyostelium

Extracellular cyclic AMP (cAMP) induces the formation of prespore cells in Dictyostelium but inhibits stalk cell formation. We have cloned gskA, which encodes the Dictyostelium homolog of glycogen synthase kinase 3 (GSK-3), and discovered that it is required for both cAMP effects. Disruption of gskA creates a mutant that aggregates but forms few spores and an abnormally high number of stalk cel...

A retinoblastoma ortholog controls stalk/spore preference in Dictyostelium.

We describe rblA, the Dictyostelium ortholog of the retinoblastoma susceptibility gene Rb. In the growth phase, rblA expression is correlated with several factors that lead to 'preference' for the spore pathway. During multicellular development, expression increases 200-fold in differentiating spores. rblA-null strains differentiate stalk cells and spores normally, but in chimeras with wild typ...
